Hands and fingers should always be relaxed. If your teacher walked up to you in class and pushed the back of your hand, your wrist should bend easily. The same goes for your fingers; if your teacher shook your hand, your fingers should move easily.
Thumbs should be relaxed which means they would be lying parallel to your palm approximately between your index and middle finger. They should never be clamped in or sticking out.
Of course there are varying moments of tension in a class, dance is fluid.
Tense hands, fingers and thumbs are a compensation for weakness in the leg, back and abdominal muscles. Being advanced too quickly leads to strain that shows up exactly where you don't want it - shouders, neck, and following that, hands, fingers and thumbs.
